If you use Direct Play on Plex, then there's no difference between that and USB.
Roku2 XS, 42" LCD, Ethernet (5.5 b320)
Roku Stick HDMI, 32" LCD, WiFi (5.5 b2027)
Roku N1000, 25" CRT, Ethernet (3.1)
Router - Linksys WRT54G2
ISP - TWC - 50 Mbps/5 Mbps
Net+ Cert

The only advantage to USB (and the reason I use for most of my local content playback on the Roku) is that all you need is the Roku and a USB drive.
Roku 2 XS>HDMI>Denon AVR-361>HDMI>Samsung 2333HD
Scientific Atlanta Cable Modem DPC2100R2 >Dynex DX-E402 Router (no wifi)
USB>Seagate Barracuda 7200 SATA 1TB in dock.

not a simple question... plex with a wired connection and a fairly descent puter is hard to beat -- wireless on an old clunker not so much....
ROKU 1 XD J0A113014862
ROKU 2 LT 16A182002191
ROKU 2 LT #2 16A19K025194
ROKU HD 18D2CP067635
all running wireless on a Netgear WNR 1000

When compared to using it with USB, which would require the content to already be in a compatible format, Plex wouldn't need to transcode, so a powerful computer isn't as important. And a wireless connections should be more than sufficient, even with the highest bitrate files that the Roku supports.

I use Plex with wifi and have had good success (good picture and sound).. Very rarely have I had a problem with rebuffering on it. Some people here don't like its menu and its setup, but I think it's easy.
Yeah, I got some Rokus.

I use both...even though Plex will play my MP4 TV shows, I prefer to load them on a flash drive & watch them via USB.
I use Plex mostly for AVI & MKV files.

I alsolike Plex for the interface on the tv. The ability to see the cover art and the synopsis of the tv show or movie. Can't get that using the usb port.

The USB channel supports artwork files....maybe not a synopsis but that hardly seems like a game changer.
